According to the tale, this organisation was created a long time ago by a Muggle King of a very far away land who wanted to keep all the magic in the world for himself. In order to do so, he needed to gather all the wizards and witches in the world, so he formed the Brigade and gave them packs of wild dogs. It is known that there was a Captain of the Brigade of Witch-Hunters, and that one of the Brigade's hounds was named Sabre and died when eating a poisonous toadstool.
When the charlatan was suspected by the King being a fraud due to Babbitty's laughing when the charlatan pretended to teach the King magic in return for gold, the charlatan blackmailed Babbitty into helping him by threatening to denouce her as a witch and hand her over to the Brigade of Witch-Hunters, whereupon she would have been torn apart by the hounds.

The Brigade was likely dissolved when the King's washerwoman, Babbitty, threatened him that he would suffer a great pain (presumably the Cruciatus Curse) if another witch or wizard was harmed because of his actions.
